Extracel™ Sponge-96 Product Data Sheet

Product Description

The Extracel Sponge-96 includes a 96-well plate pre-filled with Glycosan’s Extracel formula in a sponge format for 3-D cell culture applications. Extracel is composed of Glycosil™ (thiol-modified hyaluronic acid), Gelin-S™ (thiol-modified gelatin), and Extralink™ (polyethylene glycol diacrylate). Extracel sponges maintain their pore structure when rehydrated to provide a biocompatible scaffold for effective 3-D cell culture. 

Storage

  • Store Extracel Sponge-96 at -20 °C for up to one year.
  • Do not break the sealed packaging since this protects the product from moisture. Breaking the packaging will decrease the shelf life of the product.

Instructions for Use

Use the Extracel Sponge-96 as follows:

  1. Prepare cells for use as per standard procedures. Seeding density varies with cell type. Experimentation may be required to find the optimum seeding density for your cell culture.
  2. Prepare one 96-well plate of Extracel Sponge-96 by removing it from its packaging in aseptic conditions.
  3. Pipette 100μL of cells + media at the appropriate seeding density on top of the sponge. While pipetting, distribute the cells across the entire area of the sponge surface. If necessary, add more media to ensure that the entire sponge is thoroughly wetted. The sponge should absorb the media + cells, but there should be no excess liquid visible in the bottom of the well.
  4. Place in the 37 ˚C incubator with 5% CO2 for ½ to 1 hour to allow the cells to attach.
  5. After 1 hour, add the appropriate amount of media to the well.  Place in the 37 ˚C incubator with 5% CO2.
  6. Change the media as required using the following steps:
    • Carefully aspirate off the media from above the sponge. The sponge can be removed by the vacuum as well, so this must be done gently.
    • Add the appropriate amount of fresh media to each well without disrupting the sponge.
    • Return the plate to the 37 ˚C incubator with 5% CO2.

Note: Each sponge component has been tested and shows no evidence of bacteria growth.
