Bulk Glycosil™ Product Data Sheet

PRODUCT DESCRIPTION

Bulk Glycosil is packaged as 0.1 grams of Glycosil™ (CMHA-S, thiol-modified hyaluronic acid) per 20 mL vial. It is sterile filtered (using a 0.2 µm filter) prior to lyophilization as a 1.0 w/v% solution in 0.1x PBS. It is blanketed by argon and under a slight vacuum.

STORAGE

Glycosil™

  • Store Glycosil in original vials at -20 °C for up to one year.
  • Solutions can be stored at -20 ˚C for ~ one month if they are frozen immediately after reconstitution in their original argon blanketed vials.
    • Freeze/thaw solutions a minimum number of times for best results.
    • Solutions should be thawed at 37 °C on a rocker and used immediately after thawing.
    • During thawing, solutions go through a “gel-like” phase before returning to their liquid state.
  • Do not uncap the Glycosil vials since it will disulfide crosslink in the presence of oxygen. Use a syringe to add DG Water and remove product from the vials.

INSTRUCTIONS FOR USE

Glycosil is prepared by dissolving the lyophilized solid in the DG Water (or any sterile, degassed, deionized water of pH 5-8). When reconstituted, it will be in 0.1x phosphate buffered saline (PBS) buffer pH ~7.4 if making a 1 w/v% solution and 0.2x PBS if making a 2 w/v% solution.

Bulk Glycosil should be prepared in the following manner:

  1. Remove a Glycosil vial from the -20 ˚C freezer. Warm it to 37 ˚C (~30 minutes).
  2. Under aseptic conditions and using a syringe with the exact amount of liquid, add the following amount of DG Water to the Glycosil vial (depending on the desired final concentration)
    1. Add 5.0 mL for a 2.0 w/v% solution in 0.2x PBS
    2. Add 10.0 mL for a 1.0 w/v% solution in 0.1 x PBS
    3. Add 20.0 mL for a 0.5 w/v% solution in 0.05x PBS.
  3. Place the vial horizontally on a rocker at 37 ˚C (for maximum mixing). It will take <40 minutes for the solids to fully dissolve. The more dilute the solution, the quicker the dissolution. Solutions will be clear and slightly viscous. Note: vigorous shaking will speed up dissolving time.
  4. Use with Extralink to form a hydrogel in <30 minutes or allow to disulfide crosslink to form a hydrogel in >12 hours.

Note: Each kit component has been tested and shows no evidence of bacteria growth.
